    Observation of b2_2 symmetry vibrational levels of the SO2_2 \tilde{\mbox{C}} 1^1B2_2 state: Vibrational level staggering, Coriolis interactions, and rotation-vibration constants

    The C~\mathrm{\tilde{C}} 1^1B2_2 state of SO2_2 has a double-minimum potential in the antisymmetric stretch coordinate, such that the minimum energy geometry has nonequivalent SO bond lengths. However, low-lying levels with odd quanta of antisymmetric stretch (b2_2 vibrational symmetry) have not previously been observed because transitions into these levels from the zero-point level of the X~\mathrm{\tilde{X}} state are vibronically forbidden. We use IR-UV double resonance to observe the b2_2 vibrational levels of the C~\mathrm{\tilde{C}} state below 1600 cm−1^{-1} of vibrational excitation. This enables a direct characterization of the vibrational level staggering that results from the double-minimum potential. In addition, it allows us to deperturb the strong cc-axis Coriolis interactions between levels of a1_1 and b2_2 vibrational symmetry, and to determine accurately the vibrational dependence of the rotational constants in the distorted C~\mathrm{\tilde{C}} electronic state

    Human sperm ion channel (dys)function:implications for fertilization

    BACKGROUND: Intensive research on sperm ion channels has identified members of several ion channel families in both mouse and human sperm. Gene knock-out studies have unequivocally demonstrated the importance of the calcium and potassium conductances in sperm for fertility. In both species, the calcium current is carried by the highly complex cation channel of sperm (CatSper). In mouse sperm, the potassium current has been conclusively shown to be carried by a channel consisting of the pore forming subunit SLO3 and auxiliary subunit leucine-rich repeat-containing 52 (LRRC52). However, in human sperm it is controversial whether the pore forming subunit of the channel is composed of SLO3 and/or SLO1. Deciphering the role of the proton-specific Hv1 channel is more challenging as it is only expressed in human sperm. However, definitive evidence for a role in, and importance for, human fertility can only be determined through studies using clinical samples.OBJECTIVE AND RATIONALE: This review aims to provide insight into the role of sperm ion channels in human fertilization as evidenced from recent studies of sperm from infertile men. We also summarize the key discoveries from mouse ion channel knock-out models and contrast the properties of mouse and human CatSper and potassium currents. We detail the evidence for, and consequences of, defective ion channels in human sperm and discuss hypotheses to explain how defects arise and why affected sperm have impaired fertilization potential.SEARCH METHODS: Relevant studies were identified using PubMed and were limited to ion channels that have been characterized in mouse and human sperm. Additional notable examples from other species are included as appropriate.OUTCOMES: There are now well-documented fundamental differences between the properties of CatSper and potassium channel currents in mouse and human sperm. However, in both species, sperm lacking either channel cannot fertilize in vivo and CatSper-null sperm also fail to fertilize at IVF. Sperm-lacking potassium currents are capable of fertilizing at IVF, albeit at a much lower rate. However, additional complex and heterogeneous ion channel dysfunction has been reported in sperm from infertile men, the causes of which are unknown. Similarly, the nature of the functional impairment of affected patient sperm remains elusive. There are no reports of studies of Hv1 in human sperm from infertile men.WIDER IMPLICATIONS: Recent studies using sperm from infertile men have given new insight and critical evidence supporting the supposition that calcium and potassium conductances are essential for human fertility. However, it should be highlighted that many fundamental questions remain regarding the nature of molecular and functional defects in sperm with dysfunctional ion channels. The development and application of advanced technologies remains a necessity to progress basic and clinical research in this area, with the aim of providing effective screening methodologies to identify and develop treatments for affected men in order to help prevent failed ART cycles. Conversely, development of drugs that block calcium and/or potassium conductances in sperm is a plausible strategy for producing sperm-specific contraceptives.</p

    Strategies for Agile Leadership in a Challenging Environment

    The post pandemic environment has provided many significant challenges for all forms of organisations. The requirement for good leadership in this context is vital. This research considers the features of such leadership and how their application can affect organisations and their required outputs. The need for agility in this sphere is proposed, in order to accommodate the environmental challenges, globally and within the different sectors. There should be an emphasis on creativity in order to provide solutions to the many issues for organisations in the prevailing environment. Recruitment and retention are viewed as critical activities for sustaining these entities by ensuring the supply of the requisite standard of personnel at all levels. The latter should have suitable qualifications or the aptitude to acquire them. These staff should be given appropriate training then empowered, including being given the scope to utilise their own leadership style, as appropriate. The form of the post pandemic environment and its effects on leadership are noted. The principal factors identified here include the economic challenges, hybrid working patterns and the use of agency staff to supplement the permanent resource. The proactive development of leaders is emphasised as a critical activity for organisations, enabling them to address the issues prevalent in their environment. Research into this area occurred, using qualitative techniques. A literature review and interview with a current practitioner were performed, in order to glean the data for an inductive analysis.     Getting reticent young male participants to talk: using artefact-mediated interviews to promote discursive interaction

    During a pilot study that used interviews to collect data from young male apprentices about construction site safety, we were confronted with limited verbal responses. This article is about how we explored this research problem of ameliorating unresponsive interview interactions. The article reviews the options that previous researchers have trialled and developed, and specifically focuses on artefact-mediated interviews conducted with young male participants. We focus on the use of images within artefact-mediated interviews to draw out data from less communicative subjects. Our reflection upon this process proposes that the use of both abstract and concrete images within an artefact-mediated interview can produce diverse and enriched forms of data

    Improving safety culture: the impact of the construction induction training on the construction industry in Western Australia

    Australia strives to be a world leader in safety practice however improving safety cultures to reduce work-related injuries in the hazardous environment of the construction industry is a continual challenge. It is particularly difficult in Western Australia (WA) where the industry is engaging with the second development boom this century, often in isolated locations, and simultaneously adapting to the national harmonisation of OHS regulations. In 2006, Worksafe WA introduced mandatory certification in safety awareness training for all employees, before they could begin work on a construction site. This paper reviews the impact of this training on the construction industry, presenting both the perceptions of the stakeholders from their survey returns and through the analysis of lost time injury/disease statistics. The findings of the study indicate that the training has played a role in decreased work-related injuries and changing attitudes to safety training

    Rayleigh-B\'{e}nard convection in a homeotropically aligned nematic liquid crystal

    We report experimental results for convection near onset in a thin layer of a homeotropically aligned nematic liquid crystal heated from below as a function of the temperature difference ΔT\Delta T and the applied vertical magnetic field HH and compare them with theoretical calculations. The experiments cover the field range 8 \alt h \equiv H/ H_{F} \alt 80 (HF=H_F = is the Fr\'eedericksz field). For hh less than a codimension-two field hct≃46h_{ct} \simeq 46 the bifurcation is subcritical and oscillatory, with travelling- and standing-wave transients. Beyond hcth_{ct} the bifurcation is stationary and subcritical until a tricritical field ht=57.2h_t= 57.2 is reached, beyond which it is supercritical. The bifurcation sequence as a function of hh found in the experiment confirms the qualitative aspects of the theoretical predictions. However, the value of hcth_{ct} is about 10% higher than the predicted value and the results for kck_c are systematically below the theory by about 2% at small hh and by as much as 7% near hcth_{ct}. At hcth_{ct}, kck_c is continuous within the experimental resolution whereas the theory indicates a 7% discontinuity. The theoretical tricritical field htth=51h_t^{th} = 51 is somewhat below the experimental one. The fully developed flow above RcR_c for h<hcth < h_{ct} is chaotic. For hct<h<hth_{ct} < h < h_t the subcritical stationary bifurcation also leads to a chaotic state. The chaotic states persist upon reducing the Rayleigh number below RcR_c, i.e. the bifurcation is hysteretic. Above the tricritical field hth_t, we find a bifurcation to a time independent pattern which within our resolution is non-hysteretic.Comment: 15 pages incl. 23 eps figure

    Factors that influence career progression among postdoctoral clinical academics: a scoping review of the literature

    Background: The future of academic medicine is uncertain. Concerns regarding the future availability of qualified and willing trainee clinical academics have been raised worldwide. Of significant concern is our failure to retain postdoctoral trainee clinical academics, who are likely to be our next generation of leaders in scientific discovery. / Objectives: To review the literature about factors that may influence postdoctoral career progression in early career clinical academics. / Design: This study employed a scoping review method. Three reviewers separately assessed whether the articles found fit the inclusion criteria. / Data sources: PubMed, Scopus, Web of Science and Google Scholar (1991–2015). / Article selection: The review encompassed a broad search of English language studies published anytime up to November 2015. All articles were eligible for inclusion, including research papers employing either quantitative or qualitative methods, as well as editorials and other summary articles. / Data extraction: Data extracted from included publications were charted according to author(s), sample population, study design, key findings, country of origin and year of publication. / Results: Our review identified 6 key influences: intrinsic motivation, work–life balance, inclusiveness, work environment, mentorship and availability of funding. It also detected significant gaps within the literature about these influences. / Conclusions: Three key steps are proposed to help support postdoctoral trainee clinical academics. These focus on ensuring that researchers feel encouraged in their workplace, involved in collaborative dialogue with key stakeholders and able to access reliable information regarding their chosen career pathway. Finally, we highlight recommendations for future research

    Assortative roosting in the two phonic types of Pipistrellus pipistrellus during the mating season

    The European pipistrelle, Pipistrellus pipistrellus, consists of two phonic types that echolocate with frequencies of maximum energy (FMAXE) averaging 46 kHz and 55 kHz (named the 45 kHz and 55 kHz phonic types for simplicity). These two phonic types occur in sympatry over much of Britain. Between 1993 and 1995 mating groups of P. pipistrellus were located in bat boxes between August and October, the main mating period for these bats. Mating groups comprised a single male and up to three females. Broadband, time-expanded recordings of search-phase echolocation calls were made from bats immediately after release. The average FMAXE of each bat was calculated: bats with an FMAXE less than 49 kHz were classed as 45 kHz phonic types and bats with an FMAXE greater than 52 kHz were classed as 55 kHz phonic types. A total of 16, 45 kHz groups and ten 55 kHz groups were located. No mating group ever contained more than one phonic type, although both types were known to be present at several of the study sites. Males emitted calls of higher frequency than females for both phonic types. The study suggests that the two phonic types are reproductively isolated and supports the hypothesis that P. pipistrellus should be considered as two cryptic sibling species

    Novel pharmacological actions of Trequinsin Hydrochloride improve human sperm cell motility and function

    Background and purposeAsthenozoospermia is a leading cause of male infertility, but the development of pharmaceuticals to improve sperm motility has been hindered by the lack of effective screening platforms and knowledge of suitable molecular targets. We have demonstrated that a high throughput screening (HTS) strategy in conjunction with established in vitro tests can identify and characterise the action of compounds that improve sperm motility. The study aimed to apply HTS to identify new compounds from a novel small molecule library that increase intracellular calcium, [Ca2+]I, promote human sperm cell motility and systemically determine the mechanism of action. Experimental approach A validated HTS fluorometric [Ca2+]i assay was used to screen an in-house library of compounds. Trequinsin hydrochloride (a phosphodiesterase 3 inhibitor) was selected for detailed molecular (plate reader assays, electrophysiology and cyclic nucleotide measurement) and functional (motility and acrosome reaction) testing in sperm from healthy volunteer donors and, where possible, patients.   Key resultsThe fluorometric analysis identified Trequinsin as an efficacious agonist of [Ca2+]i, although less potent than progesterone (P4). Functionally, Trequinsin significantly increased cell hyperactivation and penetration into viscous medium in all donor sperm samples and cell hyperactivation in 22/25 (88%) patient sperm samples. The Trequinsin-induced [Ca2+]i response was cross-desensitised consistently by prostaglandin E1 but not with P4. Whole-cell patch clamp electrophysiology confirmed that Trequinsin activates CatSper and partially inhibits potassium channel activity. Trequinsin also increases intracellular cGMP.   Conclusion and Implications Trequinsin exhibits a novel pharmacological profile in human sperm and may be a suitable lead compound for the development of new pharmaceuticals to improve patient sperm function and fertilisation potential
